Characterization of a functional V1B vasopressin receptor in the male rat kidney: evidence for cross talk between V1B and V2 receptor signaling pathways
نویسندگان
چکیده
Although V 1B R mRNA has been detected in the kidney, precise renal localization as well pharmacological and physiological properties of this receptor remain unknown. Using original pharmaceutical tools, study brings new data on signaling pathways R, highlights a cross talk between 2 (V R) inner medullary collecting duct, suggests that may counterbalance some pathophysiological conditions antidiuretic effect triggered by activation.

Biological characterization of rodent and human vasopressin V1b receptors using SSR-149415, a nonpeptide V1b receptor ligand.
[(3)H]SSR-149415 is the first tritiated nonpeptide vasopressin V(1b) receptor (V(1b)R) antagonist ligand. It was used for studying rodent (mouse, rat, hamster) and human V(1b)R from native or recombinant origin. Moreover, a close comparison between the human and the mouse V(1b)R was performed using SSR-149415/[(3)H]SSR-149415 in binding and functional studies in vitro. [(3)H]SSR-149415 binding ...
متن کاملEvidence of an association between the vasopressin V1b receptor gene (AVPR1B) and childhood-onset mood disorders.
CONTEXT Disturbances in stress hormones have been implicated in mood disorders, in particular in the hyperactivity of the hypothalamic-pituitary-adrenal (HPA) axis. Arginine vasopressin (AVP) plays a crucial role in modulating the HPA axis under stress and does so through a G protein-coupled receptor, vasopressin V1b receptor (AVPR1b). OBJECTIVE To determine if genetic variation in AVPR1B cou...
متن کاملAltered lipid metabolism in vasopressin V1B receptor-deficient mice.
We previously reported that insulin sensitivity was increased in vasopressin V(1B) receptor-deficient (V(1B)R(-/-)) mice. Here, we investigate the lipid metabolism in V(1B)R(-/-) mice. Despite having lower body weight, V(1B)R(-/-) mice had significantly greater fat weight of the epididymal white adipose tissue than V(1B)R(+/+) mice. Glycerol production and beta-oxidation were suppressed in V(1B...
متن کاملPotentiation of receptor-mediated cAMP production: role in the cross-talk between vasopressin V1a and V2 receptor transduction pathways.
Cross-talk between the phospholipase C and adenylyl cyclase signalling pathways was investigated in Chinese hamster ovary (CHO) cells transfected with the V1a and V2 vasopressin receptors. Cell lines expressing V1a, V2, or both V1a and V2 receptors, were established and characterized. Stimulation of V2 receptors by vasopressin induced a dose-dependent increase in cAMP accumulation, whereas stim...
متن کاملGene and cDNA cloning and characterization of the mouse V3/V1b pituitary vasopressin receptor.
The gene of the mouse V3/V1b receptor was identified by homology cloning. One of the genomic clones contained the entire coding sequence. The cDNA presented high identity with rat (92%) and human (84%) sequences. Southern blot analysis indicated the existence of a single gene. Tissue distribution was studied by RT-PCR. The major site of expression was the pituitary. A faint signal was also pres...
